Yes — runes channeled by Virtuoso's ability enter ready.
Runes enter the board ready by default unless the effect explicitly says "channel X runes exhausted" (Rule 430.2). Since Virtuoso just says "channel 4 runes," all four come in ready. You can then use them immediately for energy that turn (FAQ #10639).
